Long-chain dicarboxylic acids play a critical role in inducing peroxisomal β-oxidation and hepatic triacylglycerol accumulation

چکیده

Recent studies provide evidence that peroxisomal β-oxidation negatively regulates mitochondrial fatty acid oxidation, and induction of causes hepatic lipid accumulation. However, whether there exists a triggering mechanism inducing is not clear. Long-chain dicarboxylic acids are the product mono subjected to ω-oxidation, both ω-oxidation induced under ketogenic conditions, indicating might be crosstalk between. Here, we revealed administration long-chain strongly induces steatosis in mice through metabolites acetyl-CoA hydrogen peroxide. Under upregulation resulted increased generation β-oxidation, which accumulation droplets animals. Inhibition reduced formation significantly lowered improved steatosis. Our results suggest endogenous act as molecules triacylglycerol deposition. Targeting an effective pathway treating liver related metabolic diseases regulating β-oxidation.

Identification of the peroxisomal β-oxidation enzymes involved in the degradation of long-chain dicarboxylic acids

Dicarboxylic acids (DCAs) are -oxidation products of monocarboxylic acids. After activation by a dicarboxylyl-CoA synthetase, the dicarboxylyl-CoA esters are shortened via -oxidation. Although it has been studied extensively where this -oxidation process takes place, the intracellular site of DCA oxidation has remained controversial. Making use of fibroblasts from patients with defined mitochon...

The catabolism of medium- and long-chain dicarboxylic acids.

The w-oxidation of fatty acids results in the formation of mediumand long-chain dicarboxylates. By this accessory metabolic pathway monocarboxylic acids are first converted into w-hydroxymonocarboxylic acids, a reaction catalysed by the microsomal mixed-oxidase function system (Pettersen, 1972). These intermediates are subsequently converted into w-oxomonocarboxylic acids and dicarboxylic acids...

Peroxisomal beta-oxidation of long-chain fatty acids possessing different extents of unsaturation.

Rates of peroxisomal beta-oxidation were measured as fatty acyl-CoA-dependent NAD+ reduction, by using solubilized peroxisomal fractions isolated from livers of rats treated with clofibrate. Medium- to long-chain saturated fatty acyl-CoA esters as well as long-chain polyunsaturated fatty acyl-CoA esters were used. Peroxisomal beta-oxidation shows optimal specificity towards long-chain polyunsat...

Hepatic triacylglycerol accumulation and insulin resistance.

The association of hepatic steatosis with hepatic insulin resistance and type 2 diabetes has prompted investigators to elucidate the underlying mechanism. In this review we focus on pathways of lipid metabolism, and we review recent data, primarily from mouse models, that link lipid intermediates with insulin resistance. Most of the studies that implicate acyl-CoA, lysophosphatidic acid, phosph...
